Defensins and Viral Infection: Dispelling Common Misconceptions
نویسندگان
چکیده
Human aand b-defensins are cationic, amphipathic effector peptides of the innate immune system with broad antimicrobial activity [1]. a-defensins are produced by neutrophils (human neutrophil peptides [HNP] 1–4), as well as by epithelial cells in the gut and genitourinary tract (human defensins [HD] 5 and 6). bdefensins (human b-defensins [HBD] 1–4) are constitutively expressed by epithelial cells of skin and mucosal surfaces. Originally discovered due to their antibacterial activity, defensins are also active against both enveloped and non-enveloped viruses. Mechanisms important for bacterial inhibition have been historically assumed to be responsible for defensin antiviral activity; however, this assumption has not held up to experimentation. Our purpose is to address some persistent myths regarding the activity of human defensins against viruses, as well as to identify areas requiring further investigation (Figure 1).

Defensins in viral infections.
Defensins are antimicrobial peptides important to innate host defense. In addition to their direct antimicrobial effect, defensins modulate immune responses. Increasing evidence indicates that defensins exhibit complex functions by positively or negatively modulating infections of both enveloped and non-enveloped viruses. The effects of defensins on viral infections appear to be specific to the...
متن کاملα-Defensin HD5 Inhibits Human Papillomavirus 16 Infection via Capsid Stabilization and Redirection to the Lysosome
α-Defensins are an important class of abundant innate immune effectors that are potently antiviral against a number of nonenveloped viral pathogens; however, a common mechanism to explain their ability to block infection by these unrelated viruses is lacking. We previously found that human defensin 5 (HD5) blocks a critical host-mediated proteolytic processing step required for human papillomav...
متن کاملCardiac misconceptions: comparisons among nurses, nursing students and people with heart disease in Taiwan.
AIM This paper is a report of a study to examine the misconceived and potentially maladaptive beliefs (cardiac misconceptions) about heart disease held by nurses, nursing students and people with heart disease in Taiwan. BACKGROUND Research suggests that misconceived and maladaptive beliefs about heart disease influence outcomes in people with heart disease, and that eliciting and dispelling ...
متن کاملAlpha-defensins block the early steps of HIV-1 infection: interference with the binding of gp120 to CD4.
Alpha-defensins are antibiotic peptides that act as natural inhibitors of HIV-1 infection. However, the mechanisms of such inhibition are still unclear. Here we demonstrate that alpha-defensins block the earliest steps in the viral infectious cycle, as documented using an HIV-1 envelope-mediated cell-fusion assay. A broad-spectrum inhibitory activity was observed on primary and laboratory-adapt...
متن کاملDirect evidence from single-cell analysis that human {alpha}-defensins block adenovirus uncoating to neutralize infection.
Human alpha-defensins are evolutionarily conserved effectors of the innate immune response with broadly acting antibacterial activity. Their role in antiviral immunity is less well understood. We previously showed that these antimicrobial peptides are potent inhibitors of human adenovirus infection. Based on biochemical studies and indirect evidence from confocal microscopy, we proposed that de...
